The shared, transdiagnostic complaint most frequently encountered in anxiety, stress, and emotion regulation disorders is insomnia. Current cognitive behavioral therapy (CBT) for these disorders often overlooks sleep, despite sleep's importance in emotional regulation and the acquisition of new cognitive and behavioral patterns, the cornerstones of CBT. This internet-delivered, guided cognitive behavioral therapy for insomnia (iCBT-I), a transdiagnostic randomized controlled trial (RCT), probes whether it (1) ameliorates sleep quality, (2) modifies the trajectory of emotional distress, and (3) amplifies the efficacy of standard treatments for emotional disorders in all mental health care (MHC) settings.
Our study targets 576 participants who manifest clinical insomnia symptoms and at least one dimension from the following diagnostic categories: generalized anxiety disorder (GAD), social anxiety disorder (SAD), panic disorder (PD), posttraumatic stress disorder (PTSD), or borderline personality disorder (BPD). Participants fall into one of three categories: pre-clinical, those without prior care, or patients referred to either general or specialized MHC facilities. Using a covariate-adaptive randomization technique, participants will be allocated to either a 5- to 8-week iCBT-I (i-Sleep) program or a control condition (sleep diary only), with follow-up assessments conducted at baseline, two months, and eight months. Substance use disorders (SUDs) might be addressed using a population-wide strategy through scalable digital therapeutic tools. Carbon dots (CDs), doped with specific elements, have garnered significant interest in cancer treatment strategies. We formulated a strategy to synthesize copper, nitrogen-doped carbon dots (Cu, N-CDs) using saffron, and then investigated their consequences for HCT-116 and HT-29 colorectal cancer (CRC) cells.
Following hydrothermal synthesis, CDs were investigated by transmission electron microscopy (TEM), energy-dispersive X-ray (EDX), Fourier transform infrared (FT-IR) spectroscopy, ultraviolet-visible (UV-Vis) absorption spectroscopy, and fluorescence spectroscopy to establish their properties. Cell viability of HCT-116 and HT-29 cells was examined after incubation with saffron, N-CDs, and Cu-N-CDs for durations of 24 and 48 hours. Cellular uptake and intracellular reactive oxygen species (ROS) were measured through the application of immunofluorescence microscopy. Lipid accumulation was monitored using Oil Red O staining. Using quantitative real-time polymerase chain reaction (q-PCR) and acridine orange/propidium iodide (AO/PI) staining, apoptosis was assessed.
